BriefDefinitive Report Human CD4 Helper T Cell Activation: Functional Involvement of Two Distinct Collagen Receptors, 1177 and VLA Integrin Family
نویسندگان
چکیده
In the present study, we showed that activation of human CD4 T cells can be induced by antiCD3 and collagen in a serum-free system . This activation was inhibited by the addition ofpeptides containing the RGD or Gly-Pro-X sequences . Significantly, we demonstrated that both the 1177 (CD26) structure and the VLA integrin family, particularly the VLA3 complex, contribute to the functional interaction between collagen and CD4 cells since anti-1177 and antiVLA3 specifically inhibited this collagen-induced CD4 cell activation . Biochemical studies showed that the 1177 structure is not a member of the VLA integrin family. These results thus indicated that two different families of antigens serve as functional collagen receptors for CD4 T cell activation.

متن کاملExpression of the α1β1 integrin, VLA-1, marks a distinct subset of human CD4+ memory T cells
The Journal of Clinical Investigation | November 2003 | Volume 112 | Number 9 Introduction The α1β1 integrin is a heterodimeric cell surface receptor composed of a β1 chain shared also by other β1 integrins and an α1 chain that contains a specific “inserted” I-domain in its extracellular portion responsible for collagen recognition (1, 2). The α1β1 integrin uniquely binds to collagen IV but als...
متن کاملIdentification of integrin collagen receptors on human melanoma cells.
Integrin receptors may mediate the adhesion of cells to a number of extracellular matrix components. We found that the attachment of human melanoma cells to collagen types I and IV was blocked by antibodies to the integrin beta 1 subunit but not by peptides containing the Arg-Gly-Asp sequence. Ligand affinity chromatography was used to search for integrin-related receptors which mediate adhesio...
متن کاملAn Alternative Leukocyte Homotypic Adhesion Mechanism, LFA-1/ICAM-l-independent, Triggered through the Human VLA-4 Integrin
The VLA-4 (CD49d/CD29) integrin is the only member of the VLA family expressed by resting lymphoid cells that has been involved in cell-cell adhesive interactions. We here describe the triggering of homotypic cell aggregation of peripheral blood T lymphocytes and myelomonocytic cells by mAbs specific for certain epitopes of the human VLAo~4 subunit. This anti-VLA-4-induced cell adhesion is isot...
متن کاملPhosphatidylinositol mannoside from Mycobacterium tuberculosis binds alpha5beta1 integrin (VLA-5) on CD4+ T cells and induces adhesion to fibronectin.
The pathological hallmark of the host response to Mycobacterium tuberculosis is the granuloma where T cells and macrophages interact with the extracellular matrix (ECM) to control the infection. Recruitment and retention of T cells within inflamed tissues depend on adhesion to the ECM. T cells use integrins to adhere to the ECM, and fibronectin (FN) is one of its major components. We have found...
